General description
The protein encoded by the human gene UBE2I (or UBC9, UBCE9) is a SUMO conjugating enzyme, also called SUMO-protein ligase, Ubiquitin carrier protein 9, Ubiquitin carrier protein I, Ubiquitin-conjugating enzyme E2 I, Ubiquitin-protein ligase I or p18, that attaches SUMO proteins (SUMO1, SUMO2, SUMO3 & SUMO4) to other target proteins, primarily various transcription factors (HIF1A, FOXL2, KAT5, I-kappa-B) with the help of ubiquitin E3 ligases such as CBX4 and RANBP2. UBE2I is also know to interact with a huge array of additional targets as part of its role as a ubiquitin E2 ligase including SALL1, FHIT, RANBP2, Protein inhibitor of activated STAT2, DNMT3A, DNMT3B, Small ubiquitin-related modifier 1, SAE2,RANGAP1,Death associated protein 6, Androgen receptor, ETS1, IPO13, TCF3, TNFRSF1A, DACH1, P53, BLMH, MAP3K1, WT1, TOP1, RAD51, Activating transcription factor 2, and PIAS1. UBE2I expression is largely nuclear and the protein can be found expressed in a wide variety of tissues including heart, skeletal muscle, pancreas, kidney, liver, lung, testis, thymus, placenta and brain. EMD-Millipore's Anti-UBE2I mouse monoclonal antibody has been tested in western blot on recombinant UBE2I protein as well as HeLa, HepG2 and COS7 cell lysates and in paraffin embedded immunohistochemistry on normal prostate and bladder cancer tissue sections. The antibody has also been successfully tested in fluorescent immunocytochemistry on HepG2 cells in culture and in flow cytometry on HepG2 cells in culture.
Immunogen
Purified recombinant fragment of human UBE2I expressed in E. Coli.
Physical form
Mouse monoclonal IgG1 ascitic fluid containing up to 0.1% sodium azide.
Unpurified
Quality
Evaluated by Western Blotting in HeLa, HepG2, and Cos7 cell lysates.
Western Blotting Analysis: A 1:500-2,000 dilution of this antibody detected UBE2I in HeLa, HepG2, and Cos7 cell lysates.
Storage and Stability
Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
